El complejo Disulfiram-cobre inhibe potentemente la actividad proteasómica en células cultivadas de cáncer de mama MDA-MB-231 y MCF10DCIS.com, pero no en células normales e inmortalizadas MCF-10A, antes de la inducción de la muerte celular apoptótica del cáncer. Este compuesto (DS), un fármaco antialcohólico de uso clínico, inhibe fuertemente la actividad constitutiva e inducida por 5-FU de NF-kappaB de manera dosis-dependiente. Inhibe tanto la translocación nuclear de NF-kappaB como la actividad de unión al ADN, pero no tiene efecto sobre la degradación de IkappaBalpha inducida por 5-FU. Este químico mejora significativamente el efecto apoptótico del 5-FU en las líneas celulares DLD-1 y RKO(WT) y potencia sinérgicamente la citotoxicidad del 5-FU en ambas líneas celulares. También anula eficazmente la quimiorresistencia al 5-FU en una línea celular resistente al 5-FU H630(5-FU) in vitro. Oseltamivir disminuye el número de células viables, y la adición de CuCl(2) mejora significativamente la muerte celular inducida por DSF a menos del 10% del control. Este compuesto administrado a células de melanoma en combinación con Cu2+ o Zn2+ disminuye la expresión de ciclina A y reduce la proliferación in vitro a concentraciones más bajas que el disulfiram solo. |

Disulfiram inhibe significativamente el crecimiento tumoral (en un 74%), asociado con la inhibición proteasómica in vivo (medida por la disminución de los niveles de actividad proteasómica del tejido tumoral y la acumulación de proteínas ubiquitinadas y sustratos proteasómicos naturales p27 y Bax) y la inducción de apoptosis (como se muestra por la activación de caspasas y la formación de núcleos apoptóticos) en ratones con xenoinjertos tumorales MDA-MB-231. Este compuesto bloquea la bomba de extrusión de P-glicoproteína, inhibe el factor de transcripción nuclear factor-kappaB, sensibiliza los tumores a la quimioterapia, reduce la angiogénesis e inhibe el crecimiento tumoral en ratones. Inhibe el crecimiento y la angiogénesis en melanomas trasplantados en ratones inmunodeficientes combinados graves, y estos efectos se potencian con la suplementación de Zn2+. |
