El tratamiento de células HCT116, SW480 y HeLa con RO-3306 durante 20 h conduce a un bloqueo completo del ciclo celular en la fase G2/M. La proliferación de HCT116 y SW480 es bloqueada eficazmente por RO-3306. RO-3306 parece ser más proapoptótico en células cancerosas (HCT116 y SW480) que en células no tumorigenicas (MCF 10A y MCF 12A). La actividad de CDK1 ciclina B1, CDK1 ciclina A, CDK2 ciclina E y CDK4 ciclina D se mide mediante un ensayo de fluorescencia resuelta en el tiempo homogénea en formato de 96 pocillos. El tampón de ensayo contenía 25 mM Hepes, 6,25 mM MgCl2, 0,003% Tween 20, 0,3 mg/mL BSA, 1,5 mM DTT y ATP de la siguiente manera: 162 μM (CDK1), 90 mM (CDK2) o 135 μM (CDK4). El tampón de CDK1 y CDK2 contenía 10 mM MgCl2. Los compuestos de prueba se diluyen en tampón de ensayo a 3 veces su concentración final en 20 μL, y la reacción se inicia añadiendo 40 μL de tampón de ensayo que contiene el sustrato pRB (0,185 μM). Las placas se incuban a 37°C durante 30 min con agitación constante, y la reacción se termina añadiendo 15 μL de anticuerpo anti-pRB fosforilado (Ser-780) a 1,6 μM en 25 mM Hepes, 24 mM EDTA y 0,2 mg/mL BSA. Tras una incubación adicional de 30 min con agitación, se añaden 15 μL de 3 nM de IgG anti-conejo marcada con Lance-Eu-W1024 y 60 nM de anticuerpo anti-His-6 conjugado con aloficocianina en 25 mM Hepes y 0,5 mg/mL BSA, y se incuba durante 1 h. Las placas se leen en el lector multi-etiquetas Victor-V con excitación a 340 nm y emisión a 615 nm y 665 nm. Los valores de IC50 se calculan a partir de las lecturas a 665 nm y se normalizan para las lecturas de Europio a 615 nm. Los valores de Ki se calculan según la ecuación: Ki = IC50/(1 + S/Km ), donde S es la concentración de ATP en el ensayo y Km es la constante de Michaelis-Menten para el ATP. La actividad inhibidora contra el panel de quinasas se determina mediante la tecnología de ensayo IMAP.
