PD 0332991 tiene poco efecto sobre otras proteínas quinasas, incluyendo EGFR, FGFR, PGFR, IR. PD 0332991 es un inhibidor no competitivo de ATP de Cdk4. PD 0332991 inhibe las células de carcinoma de mama MDA-MB-435 con una IC50 de 66 nM, lo que se debe a la reducción de la fosforilación de Rb en Ser780. Se prepara una solución madre de PD0332991 en DMSO. Los ensayos de CDK se realizan en placas de filtro de 96 pocillos. Todos los complejos quinasa CDK-ciclina se expresan en células de insecto mediante infección por baculovirus y se purifican. El sustrato para los ensayos es un fragmento (aminoácidos 792-928) de pRb fusionado a GST (GST·RB-Cterm). El volumen total en cada pocillo es de 0,1 mL que contiene una concentración final de 20 mM Tris-HCl, pH 7,4, 50 mM NaCl, 1 mM ditiotreitol, 10 mM MgCl2, 25 μM ATP (para CDK4-ciclina D1, CDK6-ciclina D2 y CDK6-ciclina D3) o 12 μM ATP (para CDK2-ciclina E, CDK2-ciclina A y CDC2-ciclina B) que contiene 0,25 μCi de [γ-32P]ATP, 20 ng de enzima, 1 μg de GST·RB-Cterm y PD 0332991 (0,001-0,1 μM). Todos los componentes, excepto el [γ-32P]ATP, se añaden a los pocillos y la placa se coloca en un mezclador de placas durante 2 min. La reacción se inicia añadiendo el [γ-32P]ATP y la placa se incuba a 25 °C durante 15 min. In vivo
El PD 0332991 indica una estasis tumoral completa en un xenoinjerto MDA-MB-435 a 150 mg/kg. El PD 0332991 también muestra una actividad antitumoral de amplio espectro en múltiples xenoinjertos tumorales humanos al eliminar el phospho-Rb y el marcador proliferativo Ki-67 del tejido tumoral y la regulación a la baja de los genes bajo el control transcripcional de E2F.
