Los ensayos de Protein Tyrosine Kinase se realizan internamente mediante métodos de ensayo basados en ELISA (IGF-1R, IR, EGFR y KDR) o en Upstate Inc. mediante un método radiométrico con ATP a una concentración de 100 µM. Los ensayos ELISA internos utilizan poli(Glu:Tyr) como sustrato unido a la superficie de placas de ensayo de 96 pocillos y la fosforilación se detecta utilizando un anticuerpo antifosfotirosina conjugado con peroxidasa de rábano. El anticuerpo unido se cuantifica utilizando ABTS como sustrato de peroxidasa midiendo la absorbancia a 405 / 490 nm. Todos los ensayos utilizan dominios catalíticos de quinasa recombinantes purificados. Las enzimas recombinantes de IGF-1R o EGFR humanos se expresan como una proteína de fusión glutatión S-transferasa NH2-terminal en células de insectos y se purifican internamente. Los valores de IC50 se determinan a partir de la curva sigmoidal dosis-respuesta del porcentaje de inhibición versus log10 de la concentración del compuesto. Linsitinib (OSI-906) inhibe el crecimiento tumoral en un modelo de ratón con xenoinjerto impulsado por IGF-1R, con un 100% de TGI y un 55% de regresión a una dosis de 75 mg/kg y un 60% de TGI y ninguna regresión a una dosis de 25 mg/kg. Su administración induce diferentes semividas de eliminación en perro, rata y ratón, que son 1,18 horas, 2,64 horas y 2,14 horas, respectivamente. Cuando se administra en diferentes dosis únicas una vez al día en ratas Sprague-Dawley hembra y ratones CD-1 hembra, revela que la Vmax no es dosis-proporcional a su dosis. Este compuesto eleva los niveles de glucosa en sangre a una dosis de 25 mg/kg después de 12 días de administración. A una dosis única de 75 mg/kg en un modelo de ratón con xenoinjerto de IGF-1R humano de longitud completa (LISN) impulsado por IGF-1R, logra una inhibición máxima de la fosforilación de IGF-1R (80%) entre 4 y 24 horas con concentraciones plasmáticas del fármaco de 26,6-4,77 μM. Cuando se administra como una dosis única de 60 mg/kg en ratones con xenoinjerto NCI-H292, inhibe la captación de glucosa a las 2, 4 y 24 horas post-tratamiento in vivo.
