SB505124 is identified as a reversible ATP competitive and selective ALK inhibitor of ALK4 and ALK5. This compound shows no toxicity to renal epithelial A498 cells at concentrations up to 100 μM for 48 hours, and blocks TGF-β–induced apoptosis of FaO cells and NRP 154 cells in a concentration-dependent manner. In human umbilical vein endothelial cells (HUVEC), this chemical (500 nM) blocks the changes of TGF-β1 on F-actin assembly and prevents ROS production induced by TGF-β. By inhibiting TGF-beta1 signaling, it leads to decreased (DFO)-induced neurogenesis. A recent study shows that this inhibitor suppresses the migration and invasion of breast cancer MCF-7-M5 cells. |

Los ensayos de quinasa se realizan como se describe por Laping et al., 2002 utilizando el dominio quinasa de ALK5 y GST-Smad3 de longitud completa fusionado en el N-terminal. Los ensayos de quinasa se realizan con 65 nM de GST-ALK5 y 184 nM de GST-Smad3 en 50 mM de HEPES, 5 mM de MgCl2, 1 mM de CaCl2, 1 mM de ditiotreitol y 3 μM de ATP. Las reacciones se incuban con 0,5 μCi de [33P]γATP durante 3 horas a 30 °C. La proteína fosforilada se captura en papel P-81, se lava con 0,5 % de ácido fosfórico y se cuenta por centelleo líquido. Alternativamente, la proteína Smad3 o Smad1 también se recubre en microplacas básicas estériles FlashPlate. Los ensayos de quinasa se realizan luego en FlashPlates con las mismas condiciones de ensayo utilizando el dominio quinasa de ALK5 con Smad3 como sustrato o el dominio quinasa de ALK6 (receptor BMP) con Smad1 como sustrato. Las placas se lavan tres veces con tampón fosfato y se cuentan con TopCount.

In a rabbit GFS model, SB505124 decreased the intraocular pressure (IOP) levels and reduces subconjunctival cell infiltration and scarring at the surgical site in the GFS. In (TAC)-treated mice and FK12EC KO mice, this compound prevents the activation of endothelial TGF-β receptors and induction of renal arteriolar hyalinosis. |
