El MK-2206 2HCl es un inhibidor alostérico y se activa por el dominio de homología de pleckstrina. Este compuesto inhibe la autofosforilación de Akt T308 y S473. También previene la fosforilación mediada por Akt de moléculas de señalización aguas abajo, incluyendo TSC2, PRAS40 y proteínas ribosomales S6. Este químico inhibe las líneas celulares Ras de tipo salvaje (WT) (A431, HCC827 y NCI-H292) más potentemente en comparación con las líneas celulares mutantes Ras (NCI-H358, NCI-H23, NCI-H1299 y Calu-6). También muestra respuestas sinérgicas en combinación con agentes citotóxicos en células tumorales pulmonares NCI-H460 u ováricas A2780. La inhibición de Akt mediada por MK-2206 o siRNA activa fuertemente la autophagy en células de glioma humano. Las quinasas Akt se ensayan mediante un sustrato peptídico biotinilado derivado de GSK. La extensión de la fosforilación del péptido se determina mediante Fluorescencia de Tiempo Resuelto Homogénea (HTRF) utilizando un anticuerpo monoclonal acoplado a un quelato de lantánido (Lance) específico para el fosfopéptido en combinación con un fluoróforo de aloficocianina unido a estreptavidina (SA-APC) que se unirá a la porción de biotina del péptido. Cuando Lance y APC están cerca, se produce una transferencia de energía no radiativa de Lance a APC, seguida de la emisión de luz de APC a 655 nm. Tampón de ensayo 10X: 500 mM HEPES, pH 7,5, 1% PEG, 16,6 mM EDTA, 1 mM EGTA, 1% BSA, 20 mM 9-glicerol fosfato; Tampón de inactivación: 50 mM HEPES pH 7,3, 16,6 mM EDTA, 0,1% BSA, 0,1% Triton X-100, 0,17 nM de anticuerpo monoclonal marcado, 0,0067 mg/mL SA-APC; Solución de trabajo de ATP/MgCl2: tampón de ensayo 1X, 1 mM DTT, 1X PIC, 5% glicerol, Akt activo; Solución de trabajo de péptido: tampón de ensayo 1X, 1 mM DTT, 1X PIC, 5% glicerol, 2 TM de péptido biotinilado GSK. La reacción se ensambla añadiendo 16 µL de solución de trabajo de ATP/MgCl2 a los pocillos apropiados. Se añade MK-2206 2HCl o vehículo (1,0 µL), seguido de 10 µL de solución de trabajo de péptido.
