Descripción Tocilizumab (anti-IL-6R) es un anticuerpo monoclonal humanizado que se une al receptor de interleucina-6, PM: 148 KD.
Objetivos

In vitro

Después de añadir tocilizumab a diferentes concentraciones en células HMO6 y CHME5 estimuladas por F3-CM o con IL6, los niveles de expresión de los marcadores M2 antiinflamatorios (CD206 y Arg1) disminuyen, así como los niveles de JAK2 y STAT3 fosforilados.
In vivo

Tocilizumab puede bloquear la activación de JAK2/STAT3 e impedir la metástasis cerebral (BM) de las células A549-F3 in vivo.

Protocolo (solo para referencia)

Ensayo celular:

  • Las células HMO6 y CHME5 con medios de control o F3-CM solos o F3-CM fueron tratadas con Tocilizumab (2.5 μg/ml durante 24 h) o con IL6 (200 ng/ml durante 24 h) solos o IL6-suplementados con Tocilizumab. Se realizó un análisis de Western blot de p-JAK2, JAK2, p-STAT3 y STAT3 en células HMO6 y CHME5.
Estudio en animales:

  • Modelos animales: Ratones desnudos de modelos de metástasis cerebral
    Dosis: 5 mg/kg
