CHIR-99021 muestra una selectividad superior a 500 veces por GSK-3 frente a sus homólogos más cercanos CDC2 y ERK2, así como otras proteínas quinasas. Además, CHIR-99021 muestra solo una unión débil a un panel de 22 receptores farmacológicamente relevantes y poca actividad inhibidora contra un panel de 23 enzimas no quinasas. CHIR-99021 induce la activación de la glucógeno sintasa (GS) en células CHO-IR que expresan el receptor de insulina con una EC50 de 0,763 μM. Además de simular las acciones de la insulina, la inhibición de GSK-3 por CHIR-99021 (3 μM) aumenta la β-catenina citosólica libre 1,9 veces, imitando la vía de señalización canónica Wnt en preadipocitos 3T3-L1. Las placas de polipropileno de 96 pocillos se llenan con 300 μL/pocillo de tampón (50 mM Tris HCl, 10 mM MgCl2, 1 mM EGTA, 1 mM ditiotreitol, 25 mM β-glicerofosfato, 1 mM NaF, 0,01% BSA, pH 7,5) que contiene 27 nM de GSK-3α o 29 nM de GSK-3β, y 0,5 μM de sustrato peptídico de biotina-CREB.
