Buparlisib (BKM120) no es sensible a las PI3K de Clase III y Clase IV ni a la PI4K. Muestra una gran actividad antiproliferativa en líneas celulares desreguladas por PI3K, incluyendo A2780, U87MG, MCF7 y DU145, con una GI50 de 0,1-0,7 nM. Este compuesto induce la apoptosis de células de mieloma múltiple (ARP1, ARK, MM.1S, MM1.R y U266), lo que resulta en un aumento de células en fase G1 y una disminución de células en fase S. También indujo la apoptosis de células primarias de MM CD138+ y tiene una citotoxicidad significativamente menor hacia las células estromales CD138-. Su exposición podría causar la regulación positiva de BimS y la regulación negativa de XIAP. BKM120 demuestra actividad antiproliferativa en líneas celulares de cáncer gástrico humano al disminuir la señalización aguas abajo de mTOR. Podría aumentar p-ERK o p-STAT3 en células de cáncer gástrico mutantes KRAS. El Buparlisib (BKM120) se disuelve en DMSO y se distribuye directamente en una placa negra de 384 pocillos a 1,25 µL por pocillo. Para iniciar la reacción, se añaden 25 µL de 10 nM de PI3 quinasa y 5 µg/mL de 1-α-fosfatidilinositol (PI) en tampón de ensayo (10 mM Tris pH 7,5, 5 mM MgCl2, 20 mM NaCl, 1 mM DTT y 0,05% CHAPS) a cada pocillo, seguido de 25 µL de 2 µM de ATP en tampón de ensayo. La reacción se realiza hasta que aproximadamente el 50% del ATP se agota y luego se detiene con la adición de 25 µL de solución KinaseGlo. La reacción detenida se incuba durante 5 minutos y el ATP restante se detecta mediante luminiscencia.

Buparlisib (BKM120) inhibe completamente pAktser473 en tumores de xenoinjerto A2780 a dosis de 30, 60 o 100 mg/kg, respectivamente, y también muestra actividad antitumoral contra el modelo de glioma U87MG a dosis de 30 y 60 mg/kg. El tratamiento con este compuesto resulta en un volumen tumoral y un nivel de cadena kappa humana circulante significativamente reducidos a 5 μM/kg/día−1 en el modelo de ratón SCID ARP1, con una supervivencia prolongada.
