SNS-032 (BMS-387032) tiene baja sensibilidad a CDK1 y CDK4 con una IC50 de 480 nM y 925 nM, respectivamente. Este compuesto mata eficazmente las células de leucemia linfocítica crónica in vitro, independientemente de los indicadores pronósticos y el historial de tratamiento. En comparación con el flavopiridol y la roscovitina, este agente es más potente, tanto en la inhibición de la síntesis de ARN como en la inducción de apoptosis. Su actividad es fácilmente reversible; la eliminación de este compuesto reactiva la ARN polimerasa II, lo que lleva a la resíntesis de Mcl-1 y a la supervivencia celular. Este inhibidor inhibe la formación de redes capilares tridimensionales de células endoteliales. Previene completamente la formación de capilares de HUVEC mediada por células U87MG. Además, este químico previene significativamente la producción de VEGF en ambas líneas celulares, previene la angiogénesis in vitro, y esta acción es atribuible al bloqueo de VEGF. Estudios preclínicos han demostrado que este compuesto induce la detención del Cell Cycle y la apoptosis en múltiples líneas celulares. Este agente bloquea el Cell Cycle mediante la inhibición de CDKs 2 y 7, y la transcripción mediante la inhibición de CDKs 7 y 9. Su actividad no se ve afectada por el suero humano. Este compuesto induce un aumento dosis-dependiente en la tinción con anexina V y la activación de caspasa-3. A nivel molecular, induce una marcada desfosforilación de serina 2 y 5 de la ARN polimerasa (ARN Pol) II e inhibe la expresión de CDK2 y CDK9 y CDK7 desfosforilada. |
