Biologische activiteit

Beschrijving Trastuzumab (anti-humaan HER2) is een gehumaniseerd, recombinant monoklonaal antilichaam dat bindt aan het extracellulaire domein van HER2, MW:145.53 KD.
Doelen
HER2
In vitro

Verlies van functie van CSK of PTEN verleent resistentie tegen zowel lapatinib als Trastuzumab in humane epidermale groeifactorreceptor 2 (HER2)-geamplificeerde maagkankercellen (GC).
In Vivo

Trastuzumab oefent antitumorale effecten uit in ERBB2-positieve AFPGC PDX-modellen, waarin statistisch significante verschillen aanwezig zijn in het tumorvolume tussen de groep behandeld met AZD5438 in combinatie met Trastuzumab en die behandeld met AZD5438 of Trastuzumab alleen in ERBB2- en CCNE1-geamplificeerde PDX-modellen.

Protocol (enkel ter referentie)

Celassay:

  • OE19-cellen getransduceerd met CSK-targeting sgRNA's of PTEN-targeting sgRNA's werden gebruikt voor de volgende test. OE19-cellen getransduceerd met niet-targeting sgRNA als controle. a Groeicurve van testgroepen met 0,05 µM lapatinib in combinatie met de aangegeven dosis Trastuzumab gedurende 6 dagen.
Dierstudie:

  • Diermodellen:Vrouwelijke BALB/c naakte muizen van ERBB2-positieve alfa-fetoproteïne producerende maagcarcinoompatiënt-afgeleide xenograftmodellen
