Biologische activiteit

Beschrijving STF-31 is een selectieve glucosetransporter GLUT1-remmer.
Doelen
GLUT1
In vitro STF-31 doodt selectief RCC's door zich specifiek te richten op glucoseopname via GLUT1. Deze verbinding remt significant de lactaatproductie en extracellulaire verzuring in VHL-deficiënte cellen met ongeveer 60%, en vermindert glycolyse door glucosetransport te verminderen. In NAPRT1-expresserende cellen vertoont het cytotoxiciteit door de enzymatische activiteit van NAMPT te remmen. Het is toxisch voor hPSC's en efficiënt voor de selectieve eliminatie van hPSC's uit gemengde culturen.
In Vivo Bij muizen met VHL-deficiënte RCC-xenografts vertraagt een beter oplosbaar analoog van STF-31 (11,6 mg/kg, i.p.) de tumorgroei aanzienlijk.

    For XTT assays, five thousand cells are plated in 96-well plates. The next day, vehicle (DMSO) or this compound is added by serial dilution. Four days later, media are aspirated, XTT solution (0.3 mg/ml of XTT, 2.65 μg/ml N-methyl dibenxopyrazine methyl sulfate in phenol red-free media) is added, and the plates are incubated at 37°C for 1-2 hours. Metabolism of XTT is quantified by measuring the absorbance at 450 nm. IC50s are calculated using linear interpolation. All conditions are measured in triplicate and each experiment is done in duplicate or triplicate.

Klantproductvalidatie

NP69 cells were transfected with LMP1 expression plasmid and treated with STF-31 or infected with lentivirus expressing shGlut-1 for 48 h. The cells were then lysed and subjected to Western blotting using specific antibodies against Glut-1 and cleaved caspase 3. β-Actin expression was used as the loading control.
