Biologische activiteit

Beschrijving LDC1267 is een zeer selectieve TAM kinase-remmer met een IC50 van <5 nM, 8 nM en 29 nM voor respectievelijk Mer, Tyro3 en Axl. Deze verbinding vertoont een lagere activiteit tegen Met, Aurora B, Lck, Src en CDK8.

In vitro LDC1267 beïnvloedt de celproliferatie matig in 11 van de 95 verschillende cellijnen met een IC50 van >5μM. In NKG2D-geactiveerde NK-cellen heft deze verbinding de remmende effecten van Gas6-stimulatie op.
In Vivo Bij B16F10 melanoom-dragende muizen verbetert LDC1267 (20 mg/kg, i.p.) efficiënt de anti-metastatische NK-celactiviteit en stoot het tumoruitzaaiingen af zonder ernstige cytotoxiciteit.

  • Kinase bindingstests

    Voor de optimalisatie van Axl/TAM receptorremmers is een Axl bindingstest ontwikkeld (HTRF-methode; Kinase tracer 236). Deze test is gebaseerd op de binding en verplaatsing van de Alexa Fluor 647-gelabelde Kinase tracer 236 aan elk glutathion S-transferase (GST)-gelabelde kinase dat in de bindingstest wordt gebruikt. Binding van de tracer aan het kinase werd gedetecteerd door gebruik te maken van europium (Eu)-gelabelde anti-GST antilichamen. Gelijktijdige binding van zowel de fluorescerende tracer als de Eu-gelabelde antilichamen aan het GST-gelabelde kinase genereert een fluorescentie resonantie energieoverdracht (FRET) signaal. Binding van de remmer aan het kinase concurreert om binding met de tracer, wat resulteert in een verlies van het FRET signaal. Voor de test wordt deze verbinding verdund in 20 mM HEPES, pH 8,0, 1 mM DTT, 10 mM MgCl2 en 0,01% Brij35. Vervolgens worden het kinase van belang (5 nM eindconcentratie), fluorescerende tracer (15 nM eindconcentratie) en LanthaScreen Eu-anti-GST antilichaam (2 nM eindconcentratie) gemengd met de respectievelijke verbindingsverdunningen (van 5 nM tot 10 μM) en gedurende 1 uur geïncubeerd. Het FRET-signaal wordt gekwantificeerd met behulp van een EnVision Multilabellreader 2104.
