In een celgebaseerde test voorkomt CH5183284 autofosforylering van FGFR1, FGFR2 en FGFR3 bij 100 tot 300 nM in de DMS114 (FGFR1-amplificatie), SNU-16 (FGFR2-amplificatie) en KMS11 [t(4;14) translocatie en FGFR3 Y373C-mutatie] cellijnen. CH5183284 produceert dus een selectieve antiproliferatieve activiteit tegen kankercellijnen die genetische afwijkingen in FGFR herbergen. CH5183284 remt ook FGFR2 dat één type van de gatekeeper-mutatie (V564F) herbergt die resistentie tegen andere FGFR-remmers veroorzaakt.
In Vivo
CH5183284 (100 mg/kg/dag, p.o.) vertoont selectieve en significante antitumoractiviteit tegen xenografts met FGFR-genetische afwijkingen zoals KG1 (leukemie, FGFR1OP-FGFR1-fusie), SNU-16 (maagkanker, FGFR2-amplificatie), MFE-280 (endometriumkanker, FGFR2 S252W-mutatie), UM-UC-14 (blaaskanker, FGFR3 S249C-mutatie) en RT112/84 (blaaskanker, FGFR3-TACC3-fusie).
De remmende activiteit van CH5183284/Debio 1347 tegen FGFR1 wordt geëvalueerd met behulp van een radiometrische filtertest door de incorporatie van 33Pi te meten met een microplaatscintillatieteller. De fosforyleringsactiviteiten van LCK, EGFR, KIT, MET, SRC, BRK, FGFR2, Flt3, LTK, INSR, YES, ABL, EPHA2, ZAP70, Fyn, IGF1R, KDR en PDGFR op substraatpeptiden worden bepaald door een homogene tijdgeresolveerde fluorescentietest met LANCE Eu-W1024-gelabeld anti-fosfotyrosine PT66-antilichaam volgens standaardmethoden. Tijdgeresolveerde fluorescentie wordt gemeten met een EnVision HTS microplaatlezer. De activiteiten van Aurora A, Akt1/PKB , PKA, Cdk1/cyclin B, Cdk2/cyclin A, PKC , PKC 1 en PKC 2 op substraatpeptiden worden bepaald door IMAP FP Screening Express Progressive Binding System. Fluorescentiepolarisatie wordt gemeten met een EnVision HTS microplaatlezer.
The cell lines are added to the wells of 96-well plates containing 0.076 to 10,000 nM CH5183284/Debio 1347 and incubated at 37°C. After 4 days of incubation, Cell Counting Kit-8 solution is added and, after incubation for several more hours, absorbance at 450 nm is measured with the iMark Microplate-Reader. The antiproliferative activity is calculated using the formula (1 − T/C) × 100 (%), where T and C represent absorbance at 450 nm of the cells treated with drugs (T) and that of untreated control cells (C). The IC50 values are calculated using Microsoft Excel 2007.
