Biologische activiteit

Beschrijving Bufalin (BF) is een krachtige remmer van steroïdreceptor-coactivator SRC-3, SRC-1 en Na+/K+-ATPase. Deze verbinding bindt aan de subeenheid α1, α2 en α3 van Na+/K+-ATPase met een Kd van respectievelijk 42,5 nM, 45 nM en 40 nM. Het is een belangrijke immunoreactieve component geïsoleerd uit het Chinese geneesmiddel Chan Su met anti-kankeractiviteit.

In vitro

Bufalin bevordert sterk de afbraak van SRC-3-eiwit en bindt direct aan de coactivator. Deze verbinding is in staat om de groei van kankercellen te blokkeren bij nanomolaire concentraties.
In Vivo

Wanneer het wordt opgenomen in een nanodeeltjesafgiftesysteem, is bufalin in staat om de tumorgroei te verminderen in een muisxenograftmodel van borstkanker. Deze verbinding maakt kankercellen gevoeliger voor andere gerichte chemotherapeutische middelen.

    Luciferase assays are performed in HeLa cells transiently transfected with the reporter vector pG5-LUC in combination with expression vectors for pBIND, pBIND SRC-1, SRC-2 or SRC-3 prior to incubation with bufalin of different doses (0, 2, 5 and 7.5 nM) for 24 hours.
